Stem cell fate depends on 'grip'

Mar. 28, 2013 ? The field of regenerative medicine holds great promise, propelled by greater understanding of how stem cells differentiate themselves into many of the body's different cell types. But clinical applications in the field have been slow to materialize, partially owing to difficulties in replicating the conditions these cells naturally experience.

A team of researchers from the University of Pennsylvania has generated new insight on how a stem cell's environment influences what type of cell a stem cell will become. They have shown that whether human mesenchymal stem cells turn into fat or bone cells depends partially on how well they can "grip" the material they are growing in.

The research was conducted by graduate student Sudhir Khetan and associate professor Jason Burdick, along with professor Christopher Chen, all of the School of Engineering and Applied Science's Department of Bioengineering. Others involved in the study include Murat Guvendiren, Wesley Legant and Daniel Cohen.

Their study was published in the journal Nature Materials.

Much research has been done on how stem cells grow on two-dimensional substrates, but comparatively little work has been done in three dimensions. Three-dimensional environments, or matrices, for stems cells have mostly been treated as simple scaffolding, rather than as a signal that influences the cells' development.

Burdick and his colleagues were interested in how these three-dimensional matrices impact mechanotransduction, which is how the cell takes information about its physical environment and translates that to chemical signaling.

"We're trying to understand how material signals can dictate stem cell response," Burdick said. "Rather than considering the material as an inert structure, it's really guiding stem cell fate and differentiation -- what kind of cells they will turn into."

The mesenchymal stem cells the researchers studied are found in bone marrow and can develop into several cell types: osteoblasts, which are found in bone; chondrocytes, which are found in cartilage; and adipocytes, which are found in fat.

The researchers cultured them in water-swollen polymer networks known as hydrogels, which share some similarities with the environments stem cells naturally grow in. These materials are generally soft and flexible -- contact lenses, for example, are a type of hydrogel -- but can vary in density and stiffness depending on the type and quantity of the bonds between the polymers. In this case, the researchers used covalently cross-linked gels, which contain irreversible chemical bonds.

When seeded on top of two-dimensional covalently cross-linked gels, mesenchymal stem cells spread and pulled on the material differently depending on how stiff it was. Critically, the mechanics guide cell fate, or the type of cells they differentiate it into. A softer environment would produce more fat-like cells and a stiffer environment, where the cells can pull on the gel harder, would produce more bone-like cells.

However, when the researchers put mesenchymal stem cells inside three-dimensional hydrogels of varying stiffness, they didn't see these kinds of changes.

"In most covalently cross-linked gels, the cells can't spread into the matrix because they can't degrade the bonds -- they all become fat cells," Burdick said. "That tells us that in 3D covalent gels the cells don't translate the mechanical information the same way they do in a 2D system."

To test this, the researchers changed the chemistry of their hydrogels so that the polymer chains were connected by a peptide that the cells could naturally degrade. They hypothesized that, as the cells spread, they would be able to get a better grip on their surrounding environment and thus be more likely to turn into bone-like cells.

In order to determine how well the cells were pulling on their environment, the researchers used a technique developed by Chen's lab called 3D traction force microscopy. This technique involves seeding the gel with microscopic beads, then tracking their location before and after a cell is removed.

"Because the gel is elastic and will relax back into its original position when you remove the cells," Chen said, "you can quantify how much the cells are pulling on the gel based on how much and which way it springs back after the cell is removed."

The results showed that the stem cells' differentiation into bone-like cells was aided by their ability to better anchor themselves into the growth environment.

"With our original experiment, we observed that the cells essentially didn't pull on the gel. They adhered to it and were viable, but we did not see bead displacement. They couldn't get a grip," Burdick said. "When we put the cells into a gel where they could degrade the bonds, we saw them spread into the matrix and deform it, displacing the beads."

As an additional test, the researchers synthesized another hydrogel. This one had the same covalent bonds that the stem cells could naturally degrade and spread through but also another type of bond that could form when exposed to light. They let the stem cells spread as before, but at the point the cells would begin to differentiate -- about a week after they were first encapsulated -- the researchers further "set" the gel by exposing it to light, forming new bonds the cells couldn't degrade.

"When we introduced these cross-links so they could no longer degrade the matrix, we saw an increase toward fat-like cells, even after letting them spread," Burdick said. "This further supports the idea that continuous degradation is needed for the cells to sense the material properties of their environment and transduce that into differentiation signals."

Burdick and his colleagues see these results as helping develop a better fundamental understanding of how to engineer tissues using stem cells.

"This is a model system for showing how the microenvironment can influence the fate of the cells," Burdick said.

The research was supported by the National Science Foundation, the National Institutes of Health and the David and Lucile Packard Foundation.

NZ's Telecom to slash over 1,000 jobs, more cuts to come

WELLINGTON (Reuters) - Telecom Corp said it will slash around 16 percent of its workforce to reduce costs by up to NZ$110 million ($92 million), the country's biggest telecommunications group said on Thursday, warning of more to come as it restructures its ailing businesses.

Telecom said it expected to cut full-time employees to 6,300 to 6,600 by mid-year, from 7,530 at the end of 2012, as the firm struggles to compete in the broadband market.

"This is an important step to build a leaner, more agile organization with a competitive cost structure, setting us up to win in the market," Telecom Chief Executive Simon Moutter said in a statement.

The restructuring, which also includes the Australian operations of its Gen-i unit, would incur a one-off cost of NZ$70 million to NZ$80 million in the current fiscal year.

Telecom said this initial exercise would cut its payroll costs by NZ$90 million to NZ$110 million on an annualized basis, pointing to more redundancy in the pipeline.

The phone operator said it is working through the remainder of its strategic change process, with further job cuts and other one-off costs expected.

"Telecom will provide a second update once decisions are taken," the company said.

The move was expected, as Telecom competes in the broadband market with Vodafone, which bought TelstraClear's operations last year.

Telecom shares were up 0.2 percent at NZ$2.325, compared with a 0.3 percent dip in the broader market.

It retained its adjusted earnings before interest, tax, depreciation and amortization guidance of NZ$1.04 billion-NZ$1.06 billion for the year, excluding the restructuring costs.

Every time we say we?ve seen everything, we manage to see something else that makes us say, ?Now we?ve seen everything.?

Pending future events, now we?ve seen everything.

A pair of 20-year-olds somehow managed to record a telephone conversation between Bills G.M. Buddy Nix and Buccaneers G.M. Mark Dominik.

Dubbed by Deadspin (which has published the audio) as ?pranksters,? there?s a chance that the state or federal authorities will apply a stronger term.

?We are aware of the report on Deadspin involving a phone conversation with Bills G.M. Buddy Nix,? the Bills told PFT? in a statement. ?We have made the league aware of the report and are reviewing it with legal counsel.?

The Buccaneers declined comment.? Per a source with knowledge of the situation, the Buccaneers likewise have referred the matter to NFL Security.

Setting aside the specific circumstances, the law generally frowns on secretly recording audio.? In many states and at the federal level, at least one party to the conversation must consent to the fact that the discussion is being taped.? Some states make it illegal even if one party to the conversation knows that the audio is being recorded.

The kids involved had no malicious intent; it was by all appearances a prank.? But if they intended to record the call (and they clearly did) and if Nix and Dominik had no idea that the call was being recorded (and they clearly didn?t), this thing could get interesting for reasons far beyond the things Nix and Dominik said to each other.

UPDATE 1:41 p.m. ET:? We won?t be discussing any of the content of the call, for now.? Because I don?t have the time to research the law and because I fear that there could be a provision extending criminal or civil liability to anyone who knowingly publishes the content of an illegally-recorded conversation, we?re being careful on this one.? (AP) ? Mohammed works at a Beirut supermarket where he waits on customers and carries their groceries home for a small tip that the 14-year-old saves to send later to his family in a village in northeastern Syria.

He is among hundreds of thousands of Syrian children who have dropped out of school and fled two years of conflict that have claimed the lives of more than 70,000 people.

He is also one of countless young Syrians now frequently seen wandering the streets of Beirut, pumping gas at stations and sometimes begging for money.

Aid groups warn that some 2 million children in Syria are facing, among other things, malnutrition, disease, early marriage and severe trauma as a result of the civil war.

To mark the second anniversary of the uprising against President Bashar Assad, the Britain-based charity Save the Children released a report Wednesday entitled "Childhood Under Fire." It says the conflict has left many children traumatized, unable to go to school and struggling to find enough to eat.

"I have to say I have been shocked and horrified by the stories that I've heard from the children here in Lebanon who fled from Syria," Justin Forsyth, chief executive of Save the Children, told The Associated Press at the group's offices in Beirut.

"You never want to hear a child talk about watching their friend killed or their father tortured in front of them or their brother shot through the leg," added Forsyth, who spent several days in Lebanon last week meeting children among the estimated 320,000 Syrian refugees who have fled to the neighboring country.

Syria's children will need decades to heal from the trauma, he warned.

Similarly, a report issued by UNICEF Tuesday said unrelenting violence, massive population displacement, and damage to infrastructure and essential services caused by the Syrian conflict risk leaving an entire generation of children scarred for life.

"As millions of children inside Syria and across the region witness their past and their futures disappear amidst the rubble and destruction of this prolonged conflict, the risk of them becoming a lost generation grows every day," said UNICEF Executive Director Anthony Lake.

The report said that in areas where the fighting is most intense, few people have access to fresh water. Also, one in five schools has been destroyed, damaged, or is being used to shelter displaced families.

In Aleppo, the center of months of fighting, only 6 percent of children are attending school, the report said.

At the same time, children are traumatized by seeing family members and friends killed and terrified by the sounds and scenes of conflict.

While the reports did not give a number of children killed or wounded in the civil war, the Violations Documentation Center in Syria, a key activist group that keeps tracks of Syria's dead, wounded and missing persons, says that some 5,500 children under the age of 15, including 3,800 boys and 1,700 girls, have been killed in the past two years.

VDC also says 901 boys and 28 girls are in detention while about 100 children are missing.

Forsyth said the 5,500 figure "is very conservative. A lot of children have been killed and injured."

Children in Syria were targeted early on in the uprising that began in March 2011, and rights groups routinely report on teenagers imprisoned and sometimes beaten and tortured.

One of the most shocking cases was that of Hamza al-Khatib, 13, who was from the southern village of Jiza in Daraa province, where the uprising first broke out after security forces arrested high school students who scrawled anti-regime graffiti on a wall.

Al-Khatib was arrested at an anti-government demonstration on April 29, 2011 and not seen again until his mutilated body, with his penis severed, was delivered to his family weeks later. Al-Khatib became a symbol of the revolt, driving thousands of protesters into the streets.

Countless other amateur videos have been posted by activists showing children who were killed by shooting, shelling or air raids. Some were only weeks-old.

In the report, it said that young boys are being used by armed groups as porters and human shields at the front lines. It added that some girls are being married off early to protect them from a widely perceived threat of sexual violence. Both sides of the conflict in Syria have accused each other of using children to protect themselves.

"The majority of people who are raped in war are usually children and that probably is the case in Syria," said Forsyth. He added that they don't have exact numbers but "I have interviewed children who were sexually harassed."

The report says that combined with the breakdown of society in parts of the country, and more than 3 million people internally displaced, the conflict has led to "the collapse of childhood for millions of youngsters."

The number of U.N.-registered refugees topped 1 million ? half of them children ? earlier this month.

Mohammed, the Beirut supermarket employee, stopped going to school after it closed because of the fighting. As the eldest of three siblings, he was sent by his family to Beirut to stay with his maternal uncle, hoping he could find work to help sustain the family.

"I make about 15,000 pounds ($10) a day," said the portly boy from the northeastern village of Shadadeh in Hassakeh province, which witnessed heavy clashes last month forcing thousands of its residents to flee.

"If I don't send money to my family, they won't be able to buy anything," he said. Mohammed gave only his first name, fearing for his security.

At a Beirut gas station, Suleiman, a teenager wearing a T-shirt and a blue baseball cap, spends his day washing cars.

"The fighting and shelling were terrifying in my city," said the boy from the oil-rich eastern city of Deir el-Zour near the border with Iraq ? an area that sees almost daily fighting between troops and rebels.

Forsyth said even though children are by nature resilient, the trauma they have been through will have a long-term impact on their lives.

"For millions of Syrian children, the innocence of childhood has been replaced by the cruel realities of trying to survive this vicious war."

(AP) ? Gonzaga played well enough to earn a No. 1 seed in the NCAA tournament, all season and in the West Coast Conference title game.

Whether the top-ranked Zags get that top spot won't be decided until the weekend, not that it matters all that much to them.

"We've been playing great basketball all year," Gonzaga forward Elias Harris said after the Zags rolled over Saint Mary's 65-51 in the WCC final Monday night. "We believe in ourselves, we believe in our coaches, we believe in our teammates."

Already headed to the NCAA tournament for the 15th straight season, Gonzaga (31-2) sure had the look of a No. 1 seed against their West Coast rivals, dominating at both ends.

The deep Zags shot 52 percent, controlled the paint and teamed up to hound Gaels leading scorer Matthew Dellavedova everywhere he went. Kelly Olynyk had 21 points and 12 rebounds, Harris added 19 points and Gonzaga had a 42-18 advantage in the paint to win its 14th straight game.

But instead of hopping up and down at their accomplishment, the Zags gave a few high-fives and hugs amid the streamers falling from the ceiling, a subdued celebration for a team that has its eyes on a bigger prize.

"You've got to enjoy your successes when they come, but you have to be short-minded in the sense that it's not the end of the year for us and we still want to come out and make some noise in the tournament," Olynyk said.

Saint Mary's (27-6) labored against Gonzaga's attacking defense and size inside, particularly after point guard Jorden Page injured his right knee midway through the first half.

Dellavedova struggled for the second straight game, scoring two points on 1-of-8 shooting, and the Gaels went 7 for 27 from 3-point range while shooting 35 percent overall.

Stephen Holt led Saint Mary's, which should have a solid enough resume to earn an at-large NCAA bid.

"I thought we played well offensively, but we didn't shoot it (well), which is a big part of offense," Saint Mary's coach Randy Bennett said.

Gonzaga has been the marker for mid-major success, reaching the NCAA tournament 14 straight seasons and the regional finals four times, including the past two seasons.

This season, the Zags broke through a big barrier on their way to 30 wins for the first time: No. 1.

Behind Olynyk, the long-haired Canadian with the Seattle-grunge vibe and way-above-the-rim game, Gonzaga lost just two games ? to ranked teams ? and became the first team to go 16-0 in the WCC. That earned the Bulldogs a spot atop The Associated Press poll for the first time last week.

While the glare at the top was too much for many teams in this season of parity, Gonzaga handled prosperity well, closing the regular season with a rout over Portland. Gonzaga then beat Loyola Marymount in the semifinals of the WCC tournament to reach the title game for the 16th straight season.

Waiting for them was a familiar foe.

The Gaels and Bulldogs have met 15 times the past five seasons, trading WCC tournament championships each of the past four years.

Gonzaga beat Saint Mary's both times they met during the regular season, 83-78 at home and 77-60 in the Bay Area. The Gaels nearly missed a third shot, needing a late 3-pointer by Dellavedova and overtime to hold off San Diego in the WCC semifinals.

Saint Mary's managed to keep it close until Page got hurt at 11:42 of first half, then went more than 6 minutes without a field goal against Gonzaga's aggressive man defense.

Olynyk brought the mostly pro-Gonzaga crowd to its feet by stripping Dellavedova and streaking in for a breakaway dunk, then Kevin Pangos kept the roar going with a jumper off another steal. "Jordy is an important part of our team and it didn't help us him going out," Dellavedova said.

Gonzaga didn't let up in the second half.

Olynyk opened with a basket inside, Bell put the Zags up 43-29 with a 3-pointer from the wing and Olynyk later put a punctuation mark on the runaway, soaring for a one-handed dunk over Brad Waldow that made it a 19-point lead.

Gonzaga rolled from there, a performance they hope is good enough for a No. 1 seed.

EA says SimCity server issues improve, not yet 'all-clear'

SAN FRANCISCO (Reuters) - Server problems that botched the launch of Electronic Arts Inc's new "SimCity" title last week and incensed gamers who could not access the game for days have improved but not yet been fully resolved, the company said.

About a week since the game's launch, the company has been scrambling to add servers and issue client updates to tackle server connectivity issues that plagued users.

While video game crashes were down 92 percent since the "SimCity" release on March 5, the company needed some more time to issue an "all-clear," Lucy Bradshaw, general manager of EA's Maxis label that develops the game, said in a blog post on Sunday.

"We need a few more days of data before we can assure you that the problem is completely solved and the game is running at 100 percent," Bradshaw said.

The rollout of the newest game in its 24-year-old "SimCity" franchise was met with an outcry on social networks and Internet forums from gamers who complained of being shut out from the game after receiving error messages.

EA said last week that it was unable to handle the user demand for its new title that is now fully hosted online and was setting up additional servers to bring the game back up.

To win back the support of its users, the company issued an apology on Friday and said it would offer them a free downloadable PC game from its portfolio.

"I know that's a little contrived - kind of like buying a present for a friend after you did something crummy. But we feel bad about what happened," Bradshaw said in a blog post on Friday.

The classic city-building game first came out in 1989 and became one of Electronic Arts' biggest hits. The new, $60 game was given a modern-day look and feel, and received positive reviews before its launch.

EA's latest, online-only "SimCity" won plaudits for incorporating some of the most topical themes in urban planning, including environmental and renewable energy issues.

Shares of Electronic Arts climbed about 4 percent to $19.24 in afternoon-trading on the Nasdaq on Monday.

(AP) ? Increased hiring, lower unemployment, stock market on the rise. Who gets the credit?

It's a hotly debated point in Washington, where political scorekeeping amounts to who gets blame and who gets praise.

Following Friday's strong jobs report ? 236,000 new jobs and unemployment dropping to a four-year low of 7.7 percent ? partisans hurriedly staked out turf.

"Woot woot!" tweeted former White House economic adviser Austan Goolsbee. "With 12 million still unemployed?" countered Senate Republican leader Mitch McConnell's spokesman, Don Stewart.

When it comes to the economy, presidents usually get the rap for downturns and reap benefits from upturns. But the main factors affecting the current recovery and the record activity in the stock market may have less to do with high-profile fiscal policy fights in Washington than they do in the decisions of the Federal Reserve Bank, which has pumped trillions of dollars into the economy, kept interests rates at near zero and pushed investors away from low-yield bonds to stocks.

"From a policy standpoint, this is being driven primarily by the Fed," said Mark Vitner, an economist at Wells Fargo.

Yet to some, Washington deserves little recognition.

"Economies recover," said Douglas Holtz-Eakin, a former director of the nonpartisan Congressional Budget Office and now head of the American Action Forum, a conservative public policy institute. He acknowledged the Fed's monetary policies halted the initial free fall by the financial industry, but he said the economy has had to catch up to the Fed's low interest rates.

"It took a long time for the housing market for them to matter and for the auto market for them to matter," Holtz-Eakin said. "So I don't think that's a policy victory."

If Democrats are eager to give President Barack Obama acclaim for spurring the recovery with an infusion of spending in 2009, there are just as many Republicans who will claim his health care law and his regulatory regimes slowed it.

If there is common ground among economists, it is that the next step in fiscal policy should be focused on reining in long-term spending on entitlements programs, particularly Medicare, instead of continuing debates over short-term spending. But such a grand bargain has been elusive, caught in a fight over Obama's desire for more tax revenue and Republican opposition to more tax increases.

Obama and some Republicans are trying to move the process with phone calls and a dinner here and a luncheon there. Next week, the president plans to address Democrats and Republicans in the House and Senate in separate meetings to see, as he put it Saturday in his weekly radio and Internet address, "if we can untangle some of the gridlock."

Who gets credit does have political consequences. A strong economy would create more space for Obama to pursue other aspects of his second-term agenda. But it's an important question for the long term, too, because if the recovery is indeed accelerating it could validate the policies that the Obama administration and the Fed put in place.

Hiring has been boosted by high corporate profits and by strength in the housing, auto, manufacturing and construction sectors. Corporate profits are up. Still, it might be too soon to declare victory. While the recovery may be getting traction, the U.S. economy is not yet strong.

Economic growth is forecast to be a modest 2 percent this year. Unemployment, even as it drops, remains high nearly four years after the end of the Great Recession, with roughly 12 million people out of work.

Last year's early months also showed strong job gains only to see them fade by June.

March could prove to be a more telling indicator as the economy responds to a third month of higher Social Security taxes and as across-the-board spending cuts that kicked in March 1 begin to work their way through government programs. Economists say anticipation of the cuts already caused a downturn in the fourth quarter of last year as the defense industry slowed spending. The Congressional Budget Office and some private forecasters say the coming cuts could reduce economic growth by about half a percentage point and cost about 700,000 jobs by the end of 2014.

"My view is that aggressive monetary and fiscal policy response to the recovery has been a net positive," said Mark Zandi, chief economist at Moody's Analytics.

But referring to the automatic cuts, he said, "Fiscal policies have turned from a very powerful tailwind to a pretty significant head wind." And, he added, "the economy is going to be tested again in the next few months."

Obama has been distancing himself from the potential consequences of the automatic cuts, even though he signed the legislation that put them in place. Initially, they were designed to be so onerous that it would force all sides to work out a long-term deficit-reduction and debt-stabilization package. But that agreement never materialized.

If the recovery has been slow, White House officials argue, it is because Republicans have been unwilling to yield to Obama's demands for deficit reduction that combines tax increases and cuts in spending.

Obama himself seemed to touch on that viewpoint in his weekly address.

"At a time when our businesses are gaining a little more traction, the last thing we should do is allow Washington politics to get in the way," he said while heralding good economic news. "You deserve better than the same political gridlock and refusal to compromise that has too often passed for serious debate over the last few years."

Vitner, the Wells Fargo economist, argues that if anyone deserves credit for the recovery, it is the American public and American businesses "for being able to tune out all the noise that's coming from Washington."

"It's remarkable," he said, "that in the face of so much political uncertainty we've been able to see the growth that we have."
